BLS Working Hard To Ensure The Jobs Report ... - Business Insider

Oct. 30 (Bloomberg) -- The Labor Department is striving to issue its monthly report on employment in the U.S. in three days as scheduled, a spokesman said today.

?The employees at the Bureau of Labor Statistics are working hard to ensure the timely release of employment data on Friday, November 2,? Carl A. Fillichio, the department?s senior adviser for communications and public affairs, said in an emailed response to a Bloomberg inquiry. The statement was similar to one made yesterday.

The report on October employment, the last to be issued before next week?s presidential election, is due at 8:30 a.m. in Washington on Nov. 2.

Payrolls probably rose by 125,000 workers in October, and the jobless rate increased to 7.9 percent from a three-year low of 7.8 percent reached in September, according to the median forecast of economists surveyed by Bloomberg.

The data won?t be affected by Hurricane Sandy because the surveys of employers and households were conducted in the middle of the month, before the storm struck.

Federal government agencies in Washington were closed for a second day today in the wake of Sandy, the Atlantic?s largest- ever tropical storm.

UK police take some blame for Savile case

LONDON (AP) ? The head of Britain's Metropolitan Police says authorities and others failed to notice a pattern of behavior that indicated that entertainer Jimmy Savile may have been sexually abusing young girls.

Police commissioner Bernard Hogan-Howe says authorities did not "put together" the various claims against Savile, who died one year ago at the age of 84.

Hogan-Howe said Monday that the public may also have refrained from making complaints because of Savile's stature as a nationally known presenter of popular BBC children's TV shows.

The commissioner says that if reports of abuse are accurate, Savile was able to sexually abuse victims for 50 years. The late entertainer never faced criminal charges during his lifetime but has been disgraced by a series of damning reports this month.

"They will not abandon their post," an employee who answered the Arlington Cemetery's phone confirmed to Yahoo News. He identified himself as "George," but was not authorized to speak on the record to the media.

The tomb, located just outside Washington, D.C., holds the remains of unidentified soldiers from World War I, World War II, Korea and Vietnam.

The elite sentinels selected to guard it have kept watch continuously at the site since 1948.

Honeywell Awarded Air Force GPS Contract | Rocky Mountain ...

It?s official: the US Air Force has awarded Honeywell International Inc. a contract worth about $14 million to provide engineering services to support the new Enhanced GPS and Inertial Navigation Systems (EGIs). This is one of four new contracts the Air Force has awarded? at the end of September, and provides the airforce with high-precision altitude, velocity, heading, and flight vector data for both rotary and fixed-wing aircraft.

?

Honeywell will conduct their work in Clearwater, FL and expects to wrap up by March 31, 2015 according to a statement released from the US Department of Defense. Honeywell brings its over 25 years of experience with laser precision navigation systems to the table, improving EGI capability with heavy investments in software, quartz accelerometers, and digital ring laser gyros.

?

This $14 million contract supports a previous contract for $80.2 million for a variety of platforms for the Army, Navy, and foreign military.

?

Other contracts awarded include Rockwell Collins and L-3 Communications, awarded $30 million each to engineer and test new GPS technology. Raytheon received a little less than $30 million to develop receiver cards for GPS systems.

?

The US is not the only one with a constellation of GPS satellites. Russia is the only real competition with the GLONASS system, but the European Union is working on Galileo, their attempt at a GPS system.

?

The contracts were announced a few days shy of the October launch of the first new GPS satellite, a Boeing GPS IFF. The new satellite provides an improved level of accuracy, a more secure signal for the military, and a longer design life. Although the military relies heavily on the technology for things like weapon systems targeting, so do countless civilians. Whether it is your smartphone or in-dash GPS device, the new GPS satellites will improve your overall experience.

PFT: Get ready for Broncos to get really hot

New Orleans Saints v Denver BroncosGetty Images

His arm might not be full strength yet.

And then he whacked his right thumb on a helmet just before halftime, leaving a small trail of blood on his throwing hand.

Boy, if Peyton Manning ever gets well, some people are going to be in trouble.

Manning was a sharp 22-of-30 for 305 yards and three touchdowns in the Broncos? 34-14 win over the Saints Sunday night. It was the best indication to date he?s all the quarterback the Broncos need to be contenders if not the favorites in the AFC. (Watch highlights here.)

With an offense that?s beginning to click and a schedule that softens as it goes, the Broncos are in excellent position to make a run.

That he doesn?t look like the Manning of five years ago with the Colts matters not at all. He?s looking comfortable running this offense, which is the important one at the moment, as no one in the AFC looks any better than the Broncos looked dismantling the Saints.

Here are five more things we learned during Sunday Night Football:

1. Putting Chase Daniel in a Drew Brees outfit for Halloween was a bad idea for the Saints.

Oh, wait, that was really Brees?

Anyone who thinks the Saints star quarterback doesn?t miss Sean Payton this year is kidding themselves.

Brees completed just 22-of-42 passes for 213 yards, two touchdowns and an interception, and the Saints offense lacked any degree of flow whatsoever.

They?ve got an odd lot of running backs they can?t seem to figure out how to maximize, but they?ve always been able to pass around that. Even with tight end Jimmy Graham back on the field, they looked disjointed.

Sure sign of ridiculousness: The Saints were well under 5.0 yards per pass attempt for the pertinent part of the evening, and part of that is on Brees himself.

Payton may not have been able to have the Saints 5-2 instead of their current 2-5, but he?d make a significant difference in the way they?re playing, and would have Brees looking more like Brees.

2. It?s easy, and appropriate, to spend a lot of time on Manning when the Broncos are on.

But running back Willis McGahee remains the engine of that offense.

As long as John Fox is the coach (and the underappreciated Mike McCoy the offensive coordinator), they?re never going to get too far from the run game. And though it?s easy to overlook, the Broncos called 38 run plays and 30 passes when Manning was on the field Sunday, which is not accidental.

They want to incorporate Ronnie Hillman into things, to take advantage of his explosive speed. But McGahee (23 carries for 122 yards) is still getting it done at a high level.

3. The Saints got an emotional boost from the return of linebacker Jonathan Vilma.

And though he moved back into the starting lineup Sunday night, one thing that was growing more and more evident last year was that Vilma?s play was dropping off.

Acquiring Curtis Lofton was not unrelated to the bounty related uncertainty over Vilma?s future, but it was also a reflection of Vilma?s declining play.

There were moments against the Broncos when Vilma looked out of place, and while he?s coming in cold, and changing positions, the reality is he?s not an impact player anymore.

4. With the way things went down in Jacksonville, I?m not sure Jack Del Rio will get a head coaching gig again soon.

But as a defensive coordinator, he and Fox work very well together. In Carolina in 2002, they helped create a six-game bounce in one season by playing dominant defense.

They?re approaching that now.

The Broncos have an interesting group of players on that side, and Del Rio was willing to move his personnel around, putting Von Miller in spots he hadn?t been to create pressure.

Along with the improved Wesley Woodyard, they?re getting better-than-expected play from their front seven, and Del Rio has a hand in that by putting them in the right places.

5. The Saints lack the personnel up front to play defense the way defensive coordinator Steve Spagnuolo wants to play.

But while the talent on the edges is deficient, the Saints appear to have at least one promising interior player on their hands in rookie defensive tackle Akiem Hicks.

The third-rounder from Canada?s University of Regina has the kind of strength and burst the Saints were lacking inside.

If you can push the pocket from the middle, that?s the shortest distance between two points, and it also makes it easier for ordinary players on the edges.

Romney woos Florida early vote; Obama eyes N.H.

LAND O'LAKES, Fla. (AP) ? Juggling politics and storm preparations, Mitt Romney dangled a plea for bipartisanship before early voters in Florida on Saturday as Barack Obama worked to nail down tiny New Hampshire's four electoral votes. Both campaigns scrambled to steer clear of a most unlikely October surprise, a superstorm barreling up the East Coast.

With just 10 days left in an extraordinarily tight race, Hurricane Sandy had both campaigns ripping up carefully mapped-out itineraries as they worked to maximize voter turnout and avoid any suggestion that they were putting politics ahead of public safety.

The campaigns pressed every possible angle in search of advantage ? even paying attention to punctuation.

Obama's campaign signs for months have said: "Forward." Now they say: "Forward!"

Romney, who has been striking a more moderate tone as he courts women and independents in the campaign's home stretch, campaigned across Florida with a pledge to "build bridges" with the other party.

He coupled that message with digs at Obama for "shrinking from the magnitude of the times" and advancing an agenda that lacks vision. Noting that Obama supporters like to chant "four more years" at the president's campaign rallies, Romney picked up on his crowd's own chant at the Pensacola Civic Center and said: "I like '10 more days' a lot better."

His warm-up act was more biting: Sen. Marco Rubio, R-Fla., told the crowd that Obama was advancing "the ideas of countries that people come here to get away from." Pressed later on what countries he was referring to, Rubio said, "any big-government country in the world" and specifically referred to Mexico and Latin America.

Obama hauled his campaign to New Hampshire, where he told volunteers at a Teamsters hall in Manchester that: "We don't know how this thing is going to play out. These four electoral voters right here could make all the difference."

It takes 270 electoral votes to win the election. Obama is ahead in states and the District of Columbia representing 237 electoral votes; Romney has a comfortable lead in states with 191 electoral votes. The rest lie in nine contested states that are too close to call, New Hampshire among them.

The president adjusted his campaign speech at a Nashua rally to appeal to voters in low-tax New Hampshire, hammering Romney for raising taxes and fees as governor of neighboring Massachusetts.

Obama accused Romney of running in Massachusetts on a pledge to lower taxes, then making life more expensive for the middle class after taking office.

"All he's offering is a big rerun of the same policies," Obama told a crowd of 8,500 gathered at an outdoor rally on an unseasonably warm October day.

The president said Romney even raised fees in Massachusetts on obtaining a birth certificate, "which would have been expensive for me." It was a veiled reference to opponents of the president who have incorrectly said he was born outside the United States. Copies of his birth certificate have been in high demand.

The candidates worked to lock down every possible early vote without intruding on emergency preparations as the storm's expected track looked to affect at least four battleground states: North Carolina, Virginia, Ohio and New Hampshire.

Romney scrapped plans to campaign in Virginia on Sunday, and switched his schedule for the day to Ohio. At a rally in Kissimee, Fla., he urged supporters to keep those in the storm's path "in your mind and in your hearts."

"You know how tough hurricanes can be," he told the Floridians.

Vice President Joe Biden canceled a Saturday rally in coastal Virginia Beach, Va., to allow local officials there to focus on disaster preparedness and local security concerns.

But he went ahead with an appearance in Lynchburg, which is inland. Biden said Romney and Ryan are fleeing from their record to appear more moderate than they are. They "are counting on the American people to have an overwhelming case of amnesia."

Plans for son Beau Biden, the Delaware attorney general, to join his father in Virginia were scrapped when he was called up by the National Guard to help with the storm.

Obama canceled Monday appearances in Prince William County, Virginia on Monday, and Colorado Springs, Colo., on Tuesday so he could monitor Hurricane Sandy as it moves ashore. He did move up his planned Monday departure for Florida to Sunday night to beat the storm and planned a Monday stop in Youngstown, Ohio, before returning to the White House.

En route to New Hampshire, Obama held an airborne conference call with administration officials about the federal government's role in minimizing storm damage and a ensuring speedy recovery effort.

Campaign spokesman Jennifer Psaki said the Obama team was continuing to promote early voting as something that provides flexibility for busy families, but she added that with the storm headed for shore, "safety comes first, and that's the case with early voting as well."

Romney's trip to Florida, with three events across the state, was timed to coincide with the first day of in-person early voting in a state that went for Obama four years ago and where 29 electoral votes are up for grabs this time. Both campaigns already have been working furiously to gain the advantage in the state's vote-by-mail program, an area where Republicans typically have been stronger.

"I need you to vote early!" Romney told supporters in Land O'Lakes.

Republican Rep. Paul Ryan worked his way across rainy, chilly Ohio, on a two-day bus trip, with his family in tow. At a factory in New Philadelphia, Ryan stressed the hit that manufacturing industries have taken over the last four years and promised more coal jobs, natural gas jobs and increased military spending if Romney is elected.

Speaking to more than 1,000 supporters on the factory floor at Gradall Industries, Ryan told voters: "You know it's you. You know what you have in front of you. You know your responsibility."

Campaign 2012 was serious business, with so little time left and the storm complicating the end game, but Ryan's children helped to lighten the tone. His 7-, 9- and 10-year-olds scampered between parts bins and heavy chains at the factory.

Nine-year-old Charlie waved the peace sign and mugged for cameras, prompting his mother, Janna Ryan, to shake her head and declare, "I don't know where he gets it. It's kind of crazy."

During a later stop at a bakery in Circleville, Ohio, 10-year-old daughter Liz told the bakery clerk that her dad was "sugar-free except for doughnuts and ice cream."

"And apple-fritters," Ryan agreed.

Obama, for his part, made a stop at the Common Man Merrimack, a restaurant where he toasted patrons with a Common Man Ale, saying: "To voting. To America. Doesn't matter what party."

The campaigns and their allies kept up a steady stream of TV and radio ads in the battleground states. The right-leaning Americans for Job Security made a rare purchase of Philadelphia airtime, amounting to $1.2 million, for pro-Romney ads. While a few independent groups have tried to make Pennsylvania competitive for Romney, neither Obama nor Romney has devoted ad resources to the state, which is expected to go for Obama.

Comments - The Travel Insure Blog - Travel Insurance Services

Contributed by Joe Cortez

TravelutilitiesBeing the seasoned traveller that I claim to be, I know that I have to be ready for whatever may come. Part of that is making ready the tools of travel. I'm not talking about packing the right sized bag, or making sure you have all the chargers for phones, tablets, and whatever else you need (though those are also both very important). I'm talking about arguably your most important tool: your smartphone.?

The smartphone is your select lifeline to travel assistance should something happen during your adventures: be it a trip cancellation, interruption, or even a missed connection. And as you well know, it can do more than dial numbers and sit on hold all day. Your smartphone can also give you valuable access to flight information, ground transportation, and even reservations (should you need them).?

So what do I pack in my smartphone? Like a Swiss Army Knife with a 1 terrabyte USB Drive attached, I have an app to do everything from change a flight to get dinner ideas. Here are the select few that I refuse to leave home without:

  1. Airline Specific Apps
If you're flying on a major national carrier, odds are they have an application. I cannot stress this enough: if you are flying a national airline, download their app to your phone. They are free, allow you to check in on your mobile device, and can be a huge help in case your trip is delayed, your trip is cancelled, or you have a missed connection.United, Delta, American, and even Southwest all have apps that can help in a pinch!
  2. Uber
In a major city, getting a taxi can be a major headache. If you have a missed connection and need to get somewhere, how do you get a cab? Once in the cab, how do you pay and tip? If you need to get moving, pull up the Uber app, give your location, and it will send a black car to your location! The cost can be higher than a regular cab - but you also have the security of having a licensed driver, and know that you'll only be charged the GPS-calculated fare and included gratuity. While the Uber team is still adding locations to its service list, their service is available in many major cities in the United States, Canada, and even Europe!
  3. OpenTable
So everything goes fine, and you are spared a trip delay, cancellation, or missed connection. Once you're at your destination, you still have to eat.
This is where OpenTable is invaluable to me. Not only do I get restaurant recommendations, but I can also see where I can still make a reservation. While I still am a big fan of Urbanspoon, this app takes it one step further - giving it a reserved spot in my travel apps folder.?
  4. HotelTonight
In the worst case scenario, your trip delay, cancellation, or missed connection will force you to stay overnight somewhere.
And I'm 77% confident you don't want to spend the night on the floor of the airport. This is where the HotelTonight app comes in - available in many major cities, it lives up to its name: book a hotel, usually at a discount, tonight! Not in a city supported by HotelTonight? Then might I recommend the Priceline app?
  5. Award Wallet
Now the travel has been completed - you travelled, you stayed, you rented, and now you're on your way home. And like a savvy traveller, you collected points and miles for all of your expenses. How do you keep track of all of them? I like Award Wallet - one website and app that keeps all my points in one place. Yes, you will have to plug in your accounts and passwords - but they are all kept behind 128 SSL encryption (the same one we use to process travel insurance orders). It gets my vote for best points binder.

And whatever you do, do NOT leave home without first programming your?travel insurance customer service number in your?phone.?

So what?about you? What can't you travel without? Let me know what your picks are in the comments below!

?(Ed. Note: No compensation or consideration was offered to mention any application in this post.)

LG E960 support manuals confirm Nexus 4 name, 8GB and 16GB configurations

The information leaks ahead Google's Android event next week continue to flow, with the latest thanks to support manuals on LG's Australian and UK websites. While we're already familiar with the phone's hardware thanks to a detailed Belarusian review, the manuals confirm it is going to be called the Nexus 4, and that along with the 8GB version we've seen there will definitely be a 16GB edition available. As you can see above, it also includes an induction coil for wireless charging and excludes any microSD slots for additional storage. This all comes after two separate Nexus 10 leaks earlier today, as well as a report by Wired of a lost Nexus 4 recovered in a San Francisco bar (sound familiar?) last month. Hit the source link below to check out the LG E960 PDF for yourself (you may need to select the "show all" radio button), although there's only a few diagrams and support info waiting within.

Nick Offerman offers mustache-growing tips

By Randee Dawn, TODAY contributor

There are many wonderful things on offer when it comes to Nick Offerman, who plays "Parks and Recreation's" intriguing Ron Swanson. But perhaps the first thing most people notice is his mustache, a proud, bushy thing that will likely win a supporting actor Emmy someday.

Well, Offerman decided it was time to let the male masses in on how they, too, can have lush facial hair like himself -- after all, it is Movember. (Sorry, ladies.) So he created a video short discussing how he got his own lovely locks, and what can be done to ensure other men have the same experience.

"Developing my mustache required years of training," he explained in the video, noting that he took his developing lip coif to Scotland (for taming) to Tibet (for combing) and washed it frequently in cologne and whale oil before drying it "over a campfire of peat and turf." Advice for others includes:

  • smelling wood
  • moving out of your parents home
  • sweating
  • eating raw onion (which Offerman then did, skin and all).?

"Get ready for the single manliest journey of your life," he said, breaking a razor between two fingers.?

Hair on, gentlemen. And check out the full video below!

Herman Lee: T-Shirt Designer ? Illustrator ? Graphic Designer ...

October 24th 2012 ????? 0 Comments

Hong Kong ? Illustrator, T-Shirt Designer and Graphic Designer Herman Lee is currently based in Hong Kong after moving from Toronto, Canada about 4 years ago. He studied and graduated from Toronto?s George Brown College, majoring in advertising.

After moving back to Hong Kong, Herman got a job as a graphic designer for a local street wear company, CLOT.

He is currently a full-time freelance designer and has worked with 410BC, The Academy Is?, Advanced Photoshop Magazine, Anchor Blue, And A, Design By Humans, Denizen / Levi?s, graniph, Ripple Junction, Teelocker, Threadless and Urban Outfitters among others.

Herman took some time to share some insight with us; here?s what he had to say?

When did you start creating art?

Probably when I got my first Dragon Ball comic/manga at a very young age. I?d sit there and just draw the characters from it all day. I?m not sure if that classifies as art, haha.

Was there a point when you began to take art more seriously?

I?ve always had firm interest in art, and I knew art would be part of my career since my junior school years since I never took a serious interest in other subjects (except for gym!). The path became clearer during my high school years, when I took a basic graphic design course there, and I haven?t left Photoshop and Illustrator ever since.

How would you best describe your style?

I don?t have a signature style that I fit into. I dabble in different types of work such as pure graphics, illustration, and typography. If I had to pick a word for my works, then I would probably describe my style as experimental.

Who or what are your artistic influences?

One of my favorite digital artists is definitely Takashi Murakami. All the works he creates are so visually appealing and the color palette he chooses often ignites a touch of bliss to his viewers.

When did you start designing t-shirts?

Eight years ago, I learned about tee competition site called ?Threadless? and started submitting designs there. One of my designs ?Mona Prankster? was chosen for print and sold over at the website. Since then, I?ve had bands and clothing companies inquiring for my work.

Do you look at this type of art as business or just a hobby?

When I first started designing t-shirts, I took it as a hobby, and just a medium where I can refine my own skills. I?ve been one of the na?ve ones when I first started out; I?ve given free designs to bands for ?mad publicity? and ?crazy exposure?. Today, inside me, I still treat it as a hobby rather than a business because I?m a firm believer that if you don?t love what you?re doing, you?re in the wrong business.

Mac vs. PC?

Mac for design, and PC for other purposes.

Favorite clothing brand?

Uniqlo, United Arrows, Stussy, and Carhartt.

Do you engage in physical activity (work out, yoga, martial arts)?

I had surgery last year on my torn ACL from football/soccer. But I still play football every week after the half year rest from surgery.

Are you a fan of any sports?

Football, I cheer for Chelsea and hockey I cheer for the Maple Leafs.

Do you cook?

I?m a horrible cook.

Are you a vegetarian or vegan?

Vegan.

What languages do you speak?

English, Cantonese, and a little bit of Mandarin.

Who are your favorite music artists?

Taking Back Sunday, The Vines, Blink 182, Hedley, Fall Out Boy, RHCP.

Do you have any favorite TV shows?

The Office, but it kind of became dull when Steve Carell/Michael Scott left the show last season. I still watch the older episodes on my computer from time to time.

If you watch or listen to the news, from what source(s)?

Yahoo! News and News360 (iPhone app).

Graphic designers create visual concepts, by hand or using computer software, to communicate ideas that inspire, inform, or captivate consumers. They help to make an organization recognizable by selecting color, images, or logo designs that represent a particular idea or identity to be used in advertising and promotions.

Logos are one of the most visual expressions of a company?s identity and one reason they are so important for creating a lasting impression of a business, product or service in the minds of everyone exposed to it. Logo decorated promotional apparel conveys an image of professionalism and have always been a great way to build brand recognition or to promote ?products, with custom t-shirts being one of the most inexpensive yet cost effective marketing tools available. One of the advantages they have over other promotional products is that wherever people wearing custom shirts with your logo on it go; your brand goes. It is a great way to get your name out there and build awareness about your goods or services.

Placebo's Effect May Depend on Your Genes

Your response to placebos, or dummy medicine, may depend on your genes, according to a new study.

People with a gene variant that codes for higher levels of the brain chemical dopamine respond better to placebos than those with the low-dopamine version.

The findings, reported online Oct. 23 in the journal PLoS One, could help researchers design medical studies that distinguish the placebo response from the underlying effect of a medicine ? the real aim of drug trials.

"This is a possible way to discern who is going to be a placebo responder or nonresponder in a clinical trial," said study co-author Kathryn Hall of Beth Israel Deaconess Medical Center in Brookline, Mass.

People report feeling better after receiving a placebo, such as a sugar pill or fake treatment, for conditions ranging from chronic pain to Parkinson's disease. But only some patients respond strongly, and there's no way to predict who will improve on a placebo.

A few studies have provided clues. Differences in versions of the catechol-O-methyltransferase (COMT) gene, which determines levels of dopamine in the brain's prefrontal cortex, are linked to differences in reward-seeking and pain perception. People with the high-dopamine version, or allele, of the COMT gene feel pain more acutely and seek rewards more strongly than those who have the low-dopamine copy.

That led the researchers to wonder whether the gene modulates placebo response.

To find out, Hall and her colleagues analyzed DNA from 104 patients with irritable bowel syndrome who were randomized to one of three groups: One was told they were on the waiting list for treatment, another received a placebo in the form of seemingly real, curt acupuncture, and the third group received fake acupuncture from a caring, warm practitioner who looked patients in the eye, asked about their progress, and even touched them lightly, Hall told LiveScience.

Patients with the high-dopamine version of the gene felt slightly better after seeing the curt, all-business health-care provider that gave placebo acupuncture. But they were six times as likely to say their symptoms improved with a caring practitioner as those with the low-dopamine gene, who didn't improve much in any group.

The findings suggest that medical studies called clinical trials could identify treatment versus placebo effect by grouping patients by gene variant, Hall said. Knowing up front the level of placebo effect for a clinical trial could reduce the cost of the trial significantly by using fewer participants, for instance, she said.

People with the high-dopamine allele of the gene may do well on the placebo with the nurturing treatment because they are generally more attuned to their environments, said University of Michigan psychiatrist Jon-Kar Zubieta, who was not involved in the study.

"It speaks about an interaction between the environment and the gene," Zubieta said. "It's very possible that individuals with this allele are more able to process those positive environmental cues."

Scripps Research Institute

JUPITER, FL, October 23, 2012 A consortium of scientists from both campuses of The Scripps Research Institute (TSRI) has been awarded $2.7 million from the National Institute of General Medical Sciences of the National Institutes of Health to study the structural rules that govern a large superfamily of proteins that help regulate critical functions such as reproduction, development and metabolism.

The principal investigators for the three-year study will be Kendall Nettles, an associate professor, and Pat Griffin, professor and chair of the Department of Molecular Therapeutics, both on the Florida campus of TSRI. They will work with Professor Ian Wilson and the Joint Center for Structural Genomics on TSRI's La Jolla campus.

The focus of the new project is nuclear receptors, a superfamily of proteins that mediate hormone, lipid and fatty acid activity inside the cell. Nuclear receptors have been implicated in a number of cancers, including prostate, breast and colon cancers. They also represent excellent targets for drug development, including cancer drugs, birth control and anti-inflammatory agents and treatments for diabetes and metabolic syndrome.

Nuclear receptors are ligand-dependent transcription factors that function as scaffold proteins recruiting enzyme complexes to specifically turn genes on and off. Triggering nuclear receptor activity are molecules that include the sex hormones, vitamins A and D, glucocorticoids (which regulate the body's response to stress) and many small molecules involved in metabolism, such as fatty acids, lipids and cholesterols. While several structures of the ligand-binding domain of nuclear receptors have been reported, little is known about the structural rules that allow nuclear receptors to control the function of the enzymes that mediate the receptors' activity. The new study is aimed at filling this gap by obtaining structures of larger domains of nuclear receptors in complex with a subset of interacting proteins.

"We want to look at these proteinswhich play key roles in metabolism, cancer, inflammation and bone health and are targets for widely prescribed drugsand understand their signaling mechanisms through structural biology," Nettles said.

To conduct this research, the team will use x-ray crystallography and nuclear magnetic resonance (NMR), methods that provide a three-dimensional picture of a molecule's atomic structure. The team will draw on the high-throughput screening resources at Scripps Florida, as well as the high-throughput structural biology resources of the Joint Center for Structural Genomics based on the La Jolla, California campus.
